Overview

HERA-CD137L (also known as HERA-4-1BBL) is a hexavalent human CD137 (4-1BB) agonist developed by Apogenix. It is engineered using Apogenix's proprietary HERA (Hexavalent TNFR-SF Receptor Agonist) technology, which consists of trivalent single-chain molecular mimics of the TNF receptor-binding domains fused to a silenced human IgG1-Fc domain dimerization scaffold. This design results in a hexavalent agonist that mimics the natural ligand-receptor binding mode, enabling high receptor clustering and activation of CD137 signaling without requiring secondary Fc receptor-mediated crosslinking. HERA-CD137L promotes anti-cancer immunity by enhancing the proliferation and cytotoxic activity of CD8+ T cells and monocytes, while leaving regulatory T (Treg) cells unaffected and preventing Treg-mediated suppression. The development of HERA-CD137L for solid tumors has been terminated.
