Overview

The herpes zoster vaccine (recombinant, adjuvanted), marketed under the brand name Shingrix, is a non-live, recombinant subunit vaccine developed by GSK for the prevention of herpes zoster (shingles) and post-herpetic neuralgia. It is indicated for adults aged 50 years and older, as well as adults aged 18 years and older who are at increased risk of herpes zoster due to immunodeficiency or immunosuppression (such as those undergoing autologous hematopoietic stem cell transplantation). The vaccine consists of recombinant varicella-zoster virus (VZV) glycoprotein E (gE), the most abundant surface glycoprotein of VZV, formulated with the liposome-based AS01B Adjuvant System (containing 3-O-desacyl-4'-monophosphoryl lipid A [MPL] and QS-21). This formulation enhances both CD4+ T-cell and humoral immune responses to prevent VZV reactivation. It is administered intramuscularly as a two-dose regimen.
