HerpesVaxTat is an experimental vaccine candidate developed by Vaxxit, currently in the IND-enabling phase of development. It utilizes a proprietary platform based on an attenuated and replication-defective Herpes Simplex Virus type 1 (HSV-1) vector that has been engineered to express the HIV-1 Tat protein. The vaccine is designed to induce a robust and broad immune response by leveraging the natural immunogenicity of the HSV-1 vector alongside the immunomodulatory properties of the Tat protein. While Tat is a key regulatory protein of HIV, in this context, it is being explored for its potential to enhance immune responses against both HSV-1 and Mycobacterium tuberculosis (TB). The replication-defective nature of the HSV-1 vector ensures that the virus cannot spread within the host, providing a favorable safety profile while allowing for the intracellular production of targeted antigens to stimulate T-cell and B-cell mediated immunity.
