HES-docetaxel is a polymer-drug conjugate consisting of the cytotoxic taxane docetaxel covalently coupled to hydroxyethyl starch (HES) via a cleavable linker. Developed by Fresenius Kabi, this conjugate is designed to improve the water solubility, tolerability, and pharmacokinetic profile of docetaxel. By utilizing HES as a macromolecular carrier, the conjugate avoids the need for irritating excipients (such as polysorbate 80) typically required to dissolve hydrophobic docetaxel, thereby reducing exposure peaks and improving safety. In preclinical models of breast cancer, HES-docetaxel demonstrated enhanced anti-tumor efficacy and superior tolerability compared to free docetaxel.
