HES6 siRNA is a small interfering RNA (siRNA) therapeutic candidate designed to target and silence the expression of Hairy and enhancer of split 6 (HES6), a transcription cofactor. Research indicates that HES6 is significantly overexpressed in malignant gliomas, such as glioblastoma multiforme (GBM) and anaplastic astrocytoma, while remaining low in normal brain tissue. In preclinical studies involving glioma cell lines (e.g., A172, LN405, U87MG), treatment with HES6 siRNA resulted in a substantial reduction in cell viability and a significant increase in apoptosis. The silencing of HES6 appears to disrupt key oncogenic pathways, affecting the expression of regulatory genes like p53, c-myc, and SP1. This approach was investigated by a consortium of Finnish research institutions, including the VTT Technical Research Centre of Finland and the University of Helsinki, as a potential strategy for treating aggressive brain tumors.
