Overview

H-DC vaccine (Heterogeneous Tumor Cell Lysate-Pulsed Dendritic Cell Vaccine) is an experimental dendritic cell-based cancer immunotherapy. It is prepared by pulsing autologous or syngeneic dendritic cells with a heterogeneous, unsorted lysate derived from bulk tumor cells, rather than a specific subpopulation such as cancer stem cells (CSCs). This vaccine is designed to present a broad spectrum of tumor-associated antigens (TAAs) to the host's immune system, thereby eliciting a multi-antigenic T-cell response involving both cytotoxic T lymphocytes (CTLs) and helper T cells. In preclinical research, particularly in studies utilizing B16-F10 melanoma and SCC7 squamous cell carcinoma models, the H-DC vaccine is frequently employed as a comparative control to evaluate the enhanced efficacy of vaccines specifically targeting CSC-enriched populations (such as ALDH-high cells). While it demonstrates some antitumor activity by targeting differentiated tumor cells, its ability to eradicate the tumor-initiating CSC population is generally limited compared to enriched CSC-DC vaccines, often resulting in higher rates of metastasis and recurrence in experimental models.
