hetIL-15-Fc (also written as hetIL-15Fc) is a preclinical recombinant fusion protein and immunotherapeutic agent consisting of a fully glycosylated heterodimeric interleukin-15 (IL-15) molecule, where the C-terminus of soluble IL-15 receptor alpha (IL-15Rα) is fused to the Fc region of human IgG1. Developed primarily by researchers at the National Cancer Institute (NCI) and National Institutes of Health (NIH), hetIL-15-Fc acts as an agonist of the IL-15 receptor complex (specifically binding to the IL-15Rβ and common γc chains on immune effector cells). The addition of the Fc fusion domain significantly increases the serum half-life and stability of the heterodimer compared to single-chain recombinant IL-15. In preclinical models, hetIL-15-Fc has demonstrated potent anti-tumor activity and robust expansion of natural killer (NK) cells and memory CD8+ T cells, showing promise for the treatment of solid tumors, including triple-negative breast cancer (TNBC), melanoma, and chronic lymphocytic leukemia.
