Hex-hR1 is a hexavalent, multivalent antibody construct targeting the insulin-like growth factor 1 receptor (IGF-IR). Developed using the proprietary Dock-and-Lock (DNL) platform technology by Immunomedics and IBC Pharmaceuticals, Hex-hR1 consists of six Fab fragments of the humanized anti-IGF-IR antibody hR1 tethered to a common Fc region. This high-valency design allows for more potent down-regulation of IGF-IR compared to the parental bivalent hR1 IgG. Preclinical studies in renal cell carcinoma (RCC) models have demonstrated that Hex-hR1 inhibits colony formation and acts synergistically with mTOR inhibitors such as temsirolimus to suppress tumor cell growth.
