hexelumab + durvalumab

Hexelumab + durvalumab is an investigational combination therapy comprising two human IgG monoclonal antibodies. Hexelumab is an anti-CD40 monoclonal antibody that acts as an agonist to augment immune responses by activating the CD40 pathway on antigen-presenting cells, thereby enhancing the priming and activation of T cells. Durvalumab is an anti-PD-L1 monoclonal antibody that blocks the interaction of PD-L1 with PD-1 and CD80; this relieves PD-L1-mediated inhibition of cytotoxic T cells and allows for enhanced immune-mediated tumor cell killing. The combination is designed to promote a more robust immune response against tumors by simultaneously activating (via CD40) and releasing inhibitory checkpoints (via PD-L1 blockade). This combination is being studied for the treatment of advanced solid tumors, including various types of cancers where such complementary immune mechanisms may be effective[2][4][5].
