HF0420 is an investigational small molecule URAT1 (Urate Transporter 1) inhibitor developed by Jiangsu Hengrui Pharmaceuticals for the treatment of hyperuricemia and gout. URAT1, encoded by the SLC22A12 gene, is a transporter protein located in the apical membrane of proximal tubule cells in the kidney, where it is responsible for the majority of uric acid reabsorption from the renal filtrate back into the blood. By selectively inhibiting URAT1, HF0420 promotes the renal excretion of uric acid, thereby effectively lowering serum urate concentrations. It is being developed as a next-generation uricosuric agent, potentially offering improved potency or a more favorable safety profile compared to existing treatments such as benzbromarone or lesinurad.
