Overview

HF1K16 is an investigational immuno-oncology drug developed by HighField Biopharmaceuticals. It is a pegylated liposome formulation encapsulating all-trans retinoic acid (ATRA), a small molecule metabolite of vitamin A. Administered by infusion, HF1K16 travels through the bloodstream and infiltrates the tumor microenvironment, where it releases ATRA to modulate immune responses. The primary mechanism involves targeting myeloid-derived suppressor cells (MDSCs), promoting their maturation into functional immune cells such as dendritic cells, which in turn activate T cell-mediated anti-tumor activity. This approach aims to relieve immune suppression within tumors and enhance anti-cancer immunity. Phase 1 clinical trials have shown that HF1K16 is safe, well-tolerated, and demonstrates preliminary efficacy in patients with refractory/metastatic solid tumors—including gliomas—where some patients achieved complete remission or prolonged stable disease[1][2][3][5][6][8].
