**HFB101110** is a humanized, afucosylated monoclonal antibody targeting **CCR8**, a chemokine receptor highly expressed on tumor-infiltrating regulatory T cells (TITRs). It exerts a dual mechanism of action: **primary** ADCC-mediated depletion of CCR8+ TITRs to reverse immunosuppression, and **secondary** blockade of CCL1 ligand binding to inhibit Treg immunosuppressive functions, chemotaxis, and calcium flux. Developed using HiFiBiO's CelliGo™ platform, it demonstrates potent binding to high- and low-copy CCR8+ cells (EC50 ~0.2-0.3 nM), specific ex vivo Treg killing from patient tumors, in vivo tumor growth delay in hCCR8 knock-in mice with Treg reduction, favorable PK (t1/2 11-15 days in cynomolgus), and good tolerability with no cytokine release or anti-drug antibodies.[1][2]
