HFE mRNA lipid nanoparticles are an experimental lipid nanoparticle (LNP)-formulated therapeutic designed to deliver mRNA encoding the HFE protein, which regulates cellular iron uptake by binding to transferrin receptor 1 (TfR1) and modulating iron homeostasis. LNPs encapsulate the mRNA to protect it from degradation, facilitate endosomal escape, and enable targeted delivery primarily to hepatocytes via apolipoprotein E (ApoE)-mediated mechanisms involving low-density lipoprotein receptor (LDLR) uptake after PEG-lipid desorption exposes ionizable lipids for protein binding. This approach addresses HFE-related disorders like hereditary hemochromatosis by restoring functional HFE expression, potentially preventing iron overload in liver and other tissues; the technology leverages ionizable cationic lipids, helper lipids (e.g., DOPE or DSPC), cholesterol, and PEG-lipids for stability and efficient cytosolic mRNA release.
