Overview

HG146 is a novel small molecule inhibitor of class I and IIb histone deacetylases (HDACs), developed by HitGen for the treatment of various cancers including adenoid cystic carcinoma, head and neck cancer, lymphoma, and multiple myeloma. Upon oral administration, it selectively inhibits the catalytic activity of class I and IIb HDACs (notably HDAC1 and HDAC2), leading to increased acetylation of chromatin histones. This results in chromatin remodeling, altered gene expression patterns—including upregulation of tumor-suppressing genes—and disruption of protein homeostasis in cancer cells to induce apoptosis. Preclinical studies have shown that HG146 has improved efficacy and safety profiles compared to existing HDAC inhibitors for multiple myeloma[1][2][5]. Clinical development is ongoing primarily in China.
