Overview

HG146 + PD-(L)1 inhibitor is a **combination investigational anti-cancer therapy** comprising HG146, a histone deacetylase inhibitor (HDACi), and a programmed cell death protein 1/ligand 1 (PD-1/PD-L1) immune checkpoint inhibitor. HG146 is administered orally, and the PD-(L)1 inhibitor component may be either a monoclonal antibody targeting PD-1 or PD-L1. This combination is designed to synergistically enhance anti-tumor immunity: HG146 increases T-cell recruitment by inducing expression of chemokines (such as Ccl5, Cxcl9, and Cxcl10) and may sensitize tumors to checkpoint blockade by modulating epigenetic and immune pathways. The PD-(L)1 inhibitor blocks immune checkpoint signaling to reinvigorate exhausted T cells and enable immune-mediated clearance of cancer cells. The combination is being investigated in early-phase clinical trials for patients with refractory/relapsed solid tumors and lymphomas, with expectation to overcome immune resistance and improve clinical responses relative to single-agent therapy[2].
