HG153 is a small molecule inhibitor of the menin-MLL protein-protein interaction, developed by HitGen and Yunzhong Pharmaceutical. It targets the interaction between menin and the N-terminus of MLL (KMT2A), which is essential for the maintenance of HOXA9 and MEIS1 gene expression in certain leukemias. By disrupting this complex, HG153 induces cell cycle arrest, differentiation, and apoptosis in MLL-rearranged and NPM1-mutant leukemia cells. The drug is formulated as an oral tablet and is currently in preclinical development for the treatment of acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL) harboring these specific genetic alterations.
