HG381 is a small molecule agonist of the **Stimulator of Interferon Genes (STING)** protein, developed by **HitGen** for the treatment of advanced solid tumors. STING is a critical mediator of the innate immune system; its activation leads to the induction of Type I interferons and pro-inflammatory cytokines, which in turn promote the recruitment and activation of T cells and natural killer (NK) cells against tumor antigens. Unlike many early-generation STING agonists that required intratumoral injection, HG381 is designed for systemic administration via **intravenous** infusion, potentially allowing it to target both primary tumors and distant metastases. The compound was discovered using HitGen's DNA-encoded library (DEL) platform and is currently undergoing Phase 1 clinical evaluation to determine its safety, tolerability, and preliminary anti-tumor activity in patients with refractory solid malignancies.
