An HGF-specific antibody (or anti-HGF antibody) is a class of therapeutic monoclonal antibodies designed to target and neutralize hepatocyte growth factor (HGF), a pleiotropic cytokine and the sole ligand for the c-Met receptor tyrosine kinase. By binding to HGF, these antibodies prevent its interaction with c-Met, thereby inhibiting downstream signaling pathways (such as PI3K/Akt, MAPK, and STAT3) that drive tumor cell proliferation, survival, motility, invasion, and angiogenesis. This pathway is also implicated as a key escape mechanism and driver of resistance to epidermal growth factor receptor (EGFR) inhibitors. Prominent clinical-stage candidates in this class include ficlatuzumab (AV-299), which is currently in Phase 3 development for head and neck squamous cell carcinoma (HNSCC), and rilotumumab (AMG 102), whose development in gastric cancer was terminated due to safety concerns.
