HGFK1 is the **kringle 1 domain of the human hepatocyte growth factor alpha chain (HGFα)**, a recombinant protein or gene therapy candidate with notable *anti-angiogenic* and *anti-tumor* properties. It acts as an antagonist of the hepatocyte growth factor (HGF)/MET signaling pathway, blocking tumor neovascularization, suppressing cancer stem cell (CSC) populations (particularly CD90+ CSCs in hepatocellular carcinoma), and inhibiting key signaling pathways such as Wnt/β-catenin, Notch, and pre-MET. HGFK1 demonstrates synergistic effects with other anti-angiogenic agents like Endostatin and chemotherapeutics like cisplatin, promoting cancer cell differentiation and chemosensitivity[1][2][3][4][5]. It has potential applications in hepatocellular carcinoma, breast cancer (where it inhibits bone metastasis by activating TAK1/p38 MAPK and inhibiting RANK), renal cell carcinoma, non-small cell lung cancer, and glioblastoma. Delivery formats include recombinant protein (rHGFK1), plasmid-based gene therapy, and nanoparticle-based systems.
