hGGP (also known as LARISSA in clinical trial contexts) is a recombinant live-attenuated vaccine candidate designed for the prevention of Rift Valley fever (RVF), a zoonotic disease caused by the Rift Valley fever virus (RVFV). Developed by Wageningen University & Research and progressed into clinical development by Cr2o B.V., the vaccine utilizes a modified version of the virus where the glycoproteins (Gn and Gc) and the p78 protein are expressed from human-codon-optimized sequences (hGGP). This optimization is intended to enhance protein expression in human cells, thereby improving the safety and immunogenicity profile compared to traditional attenuated strains. The vaccine is designed for single-dose administration to provide robust, long-lasting immunity. It is currently being evaluated in a Phase 1 first-in-human study (LARISSA trial) to assess its safety, tolerability, and immunogenicity in healthy adults.
