hgp100 peptide is a synthetic peptide vaccine consisting of the 25-33 amino acid sequence (KVPRNQDWL) of the human melanoma-associated antigen glycoprotein 100 (gp100). It serves as an altered peptide ligand that exhibits higher affinity for the T-cell receptor (TCR) of gp100-specific CD8+ T cells compared to the native murine sequence, making it a potent tool for inducing antitumor immune responses in preclinical models and clinical settings. The peptide is typically administered to prime and expand cytotoxic T lymphocytes (CTLs) that can recognize and lyse melanoma cells expressing the gp100 antigen. It is frequently studied in combination with immunomodulatory agents, such as CD27 agonists (e.g., varlilumab) and PD-1/PD-L1 checkpoint inhibitors, to overcome immunosuppression and enhance the efficacy of cancer immunotherapy.
