Overview

HGR4113 is a novel small molecule insulin sensitizer developed by Glaceum Inc. for the treatment of type 2 diabetes mellitus (T2DM). Discovered through a Quantitative Structure-Activity Relationship (QSAR) study of glabridin (a natural compound from licorice) and its derivatives, HGR4113 is designed to provide potent antihyperglycemic effects comparable to thiazolidinediones (TZDs) like pioglitazone, but without the associated adverse effect of significant weight gain. In preclinical studies using db/db mice, HGR4113 demonstrated a level of efficacy in reducing fasting glucose and HbA1c levels highly comparable to pioglitazone while maintaining a body weight profile similar to metformin. The compound has progressed into Phase 1 clinical trials to evaluate its safety, tolerability, and pharmacokinetic profile following single and multiple oral doses in healthy subjects.
