Overview

Conformation-X Therapeutics is developing a first-in-class HHLA2 (Human Endogenous Retrovirus-H Long Terminal Repeat-Associating 2) inhibitor program for the treatment of cancer. HHLA2 is an immune checkpoint protein belonging to the B7 family that is frequently overexpressed in various solid tumors, particularly those that are PD-1/PD-L1 negative, where it facilitates tumor immune evasion by inhibiting T-cell proliferation and cytokine production. The program utilizes the Pledge Therapeutics discovery platform to develop both first-in-class biologics and orally available small molecule drugs designed to selectively block HHLA2's immunosuppressive effects. In April 2024, the company announced successful ex vivo and in vivo proof-of-concept studies demonstrating the program's ability to restore anti-tumor immune responses, positioning it as a potential therapy for cancers resistant to current checkpoint inhibitors.
