Hi-8 MEL was an investigational **heterologous prime-boost therapeutic cancer vaccine** for HLA-A2-positive advanced, nonresectable melanoma. It used a plasmid-DNA prime followed by a recombinant modified vaccinia Ankara boost, with both components encoding the Mel3 polyepitope string containing seven HLA-A2/A1-restricted epitopes from five melanoma-associated antigens: Melan-A, MAGE-1, MAGE-3, NY-ESO-1, and tyrosinase. The regimen was designed to prime and amplify antigen-specific cytotoxic CD8-positive T-cell responses against melanoma cells. Oxxon Therapeutics completed a Phase II study; Oxford BioMedica subsequently acquired Oxxon and later wrote off the Hi8 MEL asset after an unsuccessful divestment process.
