Overview

hI-con1 is an antibody-like immunoconjugate molecule designed to target tissue factor (TF), a protein overexpressed in certain cancers. The drug consists of two human Factor VII (fVII) domains fused to the Fc region of human immunoglobulin G1 (IgG1). This structure allows hI-con1 to bind specifically and with high affinity to tissue factor on tumor cells, facilitating immune-mediated cytotoxicity against these cells. It has been primarily investigated for use in chemotherapy-resistant uterine serous papillary adenocarcinoma (USPC), where it demonstrated strong cytotoxic effects against TF-expressing cancer cell lines. The addition of low-dose interleukin-2 further enhances its cytotoxic activity. Developed initially by Yale University, hI-con1 represents a novel targeted therapy approach for advanced or refractory cancers expressing tissue factor[3][2][1].
