Overview

Hi-TCR-T is a multi-target T cell receptor-engineered T cell therapy (TCR-T) under clinical investigation for the treatment of advanced hepatocellular carcinoma. This therapy involves collecting peripheral blood lymphocytes from patients, engineering them ex vivo to express high-affinity T cell receptors targeting multiple tumor-associated antigens (including FAP), and then reinfusing these modified cells back into the patient following lymphodepleting chemotherapy with fludarabine and cyclophosphamide. The goal is to enhance the immune system’s ability to recognize and destroy cancer cells by targeting intracellular antigens presented on tumor cells via major histocompatibility complex (MHC) molecules. The approach allows for customization of target combinations or dosing based on disease progression, as evaluated by a multidisciplinary team[6].
