Overview

Hi-TCR-T cell injection is an autologous T-cell receptor (TCR) engineered T-cell therapy developed by Shanghai Sixth People's Hospital for the treatment of advanced solid tumors, including osteosarcoma and small cell lung cancer. The therapy utilizes a multi-antigen targeting approach, directed against tumor-associated antigens such as Nectin-4, NKG2D ligands (NKG2DL), TROP2, B7-H3, and Fibroblast Activation Protein (FAP). To enhance anti-tumor activity and overcome the immunosuppressive tumor microenvironment, the cells are engineered to secrete a cocktail of immunostimulatory factors, including Interleukin-7 (IL-7), a super-agonist version of Interleukin-15 (super IL-15), and the chemokine CCL21. Additionally, the cells express single-chain variable fragments (scFvs) targeting TGF-beta1 and PD-1 to block inhibitory signaling pathways. The "Hi" in Hi-TCR-T stands for "Highly-integrated," reflecting the complex genetic engineering involved in combining multiple targeting and regulatory elements into a single therapeutic product.
