Drug intelligence / Profile preview
HiaBD1 is a recombinant protein subunit vaccine candidate under development by the University of Pennsylvania and the Children's Hospital of Philadelphia. It consists of the highly conserved binding domain 1 (BD1) of the Haemophilus influenzae adhesin (Hia), a surface-exposed trimeric autotransporter protein that mediates high-affinity adherence to human respiratory epithelial cells. While the passenger domain of Hia exhibits sequence variability, the HiaBD1 domain is highly conserved across diverse strains of Nontypable Haemophilus influenzae (NTHi). In preclinical studies, intranasal immunization with recombinant HiaBD1 formulated with the mucosal adjuvant dmLT (double-mutant heat-labile toxin) elicited robust systemic and mucosal antibody responses and significantly reduced nasal bacterial burdens in mice, demonstrating its potential to prevent NTHi-mediated respiratory tract infections.
