Overview

HIF-1α shRNA is an experimental RNA interference (RNAi) construct designed to silence the expression of the Hypoxia-Inducible Factor-1 alpha (HIF-1α) protein by targeting its encoding gene, *HIF1A*. HIF-1α is a critical transcription factor that regulates the cellular response to hypoxia and is frequently overexpressed in solid tumors, where it promotes glycolysis, angiogenesis, metastasis, and resistance to therapy. By utilizing short hairpin RNA (shRNA) technology, typically delivered via viral vectors (such as lentivirus) or plasmids, researchers can achieve stable knockdown of HIF-1α to study its role in tumor biology and evaluate its potential as a therapeutic target. Preclinical studies have explored its use in various cancers, including pancreatic, gastric, and hepatocellular carcinoma, often demonstrating reduced tumor growth and increased sensitivity to chemotherapy or embolization.
