HIF-2α siRNA (small interfering RNA targeting hypoxia-inducible factor-2alpha, also known as EPAS1) is a class of RNA interference (RNAi) therapeutics designed to silence the expression of the EPAS1 gene. HIF-2α is a key transcription factor that is stabilized under hypoxic conditions and plays a critical role in regulating cellular responses to low oxygen, including angiogenesis, cell proliferation, and metastasis. In oncology, particularly in clear cell renal cell carcinoma (ccRCC) where the von Hippel-Lindau (VHL) tumor suppressor is frequently inactivated, HIF-2α is constitutively active and drives tumor progression. Silencing HIF-2α via siRNA leads to the degradation of EPAS1 mRNA, thereby downregulating downstream oncogenic targets such as VEGF. Beyond oncology, HIF-2α siRNA has been investigated in preclinical models for inflammatory and degenerative diseases, such as osteoarthritis, where HIF-2α acts as a catabolic regulator of cartilage destruction.
