HIF1-alpha-specific siRNA is a type of small interfering RNA (siRNA) designed to selectively silence the expression of the Hypoxia-inducible factor 1-alpha (HIF1A) gene. HIF1-alpha is a critical transcription factor that regulates the cellular response to hypoxia, a condition frequently found in the microenvironment of solid tumors like glioblastoma multiforme (GBM). By facilitating the degradation of HIF1A mRNA through the RNA-induced silencing complex (RISC), this siRNA reduces the synthesis of HIF1-alpha protein. This inhibition leads to the downregulation of various downstream targets involved in tumor progression, including those related to angiogenesis, metabolic reprogramming, and the maintenance of cancer stem cell populations (e.g., CD133). In research settings, it is utilized to investigate the role of hypoxia in tumor resistance and to evaluate the potential of HIF1-alpha as a therapeutic target.
