Overview

High-dose interferon (HDI) refers to the use of large doses of the cytokine drug interferon alfa-2b, most commonly in the adjuvant treatment of melanoma. Interferons are naturally occurring proteins with antiviral, antiproliferative, and immunomodulatory effects. In oncology, HDI is used to stimulate immune responses against tumor cells. The mechanism involves binding to cell surface receptors and activating intracellular signaling pathways that enhance immune surveillance and inhibit tumor proliferation. High-dose regimens are associated with significant toxicity but have been shown to improve relapse-free survival in certain populations with high-risk melanoma.
