Overview

High-molecular-weight formalinized allergoids are chemically modified allergen extracts created by treating natural allergens with formaldehyde (formalin), resulting in covalent cross-linking and polymerization of the proteins. This process produces large molecular weight complexes (typically above 85,000 Da and up to 10^5–10^6 Da) that retain immunogenicity but have significantly reduced IgE-mediated allergenicity. The modification allows for safer administration of higher doses during allergen immunotherapy (AIT), reducing the risk of systemic allergic reactions while maintaining or enhancing the ability to induce immune tolerance through increased production of blocking antibodies and regulatory T cell responses. These preparations are used primarily in subcutaneous or sublingual immunotherapy for allergic diseases such as allergic rhinoconjunctivitis and grass pollen allergy[3][4][5][6].
