High-purity human natural killer (NK) cells are an investigational adoptive cell therapy developed by The Affiliated Cancer Hospital of Guizhou Medical University for the treatment of hematological malignancies, specifically relapsed or refractory diffuse large B-cell lymphoma (DLBCL). This therapy involves the isolation, expansion, and infusion of highly purified NK cells, which are innate lymphoid cells capable of exerting potent anti-tumor activity without the need for prior antigen sensitization. The mechanism of action involves the recognition of tumor cells through activating receptors such as NKG2D and CD16, followed by the release of cytotoxic granules containing perforin and granzymes that induce target cell apoptosis. Additionally, these NK cells secrete pro-inflammatory cytokines like interferon-gamma (IFN-γ) to enhance the overall anti-tumor immune response. The therapy is currently being evaluated in early-phase clinical trials to establish its safety profile and therapeutic potential in patients who have failed standard-of-care treatments.
