HIKESHI shRNA is a short hairpin RNA-based therapeutic candidate designed to suppress the expression of HIKESHI, a nuclear transporter protein. HIKESHI is responsible for transporting molecular chaperones, specifically HSP70 and HSP110/105, into the nucleus under heat stress conditions to prevent cancer cell death. In pancreatic cancer, HIKESHI is often overexpressed and serves as an independent poor prognostic marker for overall survival. By silencing HIKESHI, this shRNA inhibits the nuclear translocation of these heat shock proteins, leading to reduced cell viability, inhibited colony formation, and suppressed tumor growth in preclinical models. The research is primarily led by investigators at Gunma University, who have demonstrated its efficacy in both in vitro and in vivo pancreatic cancer models.
