Overview

HIL-214 (formerly TAK-214) is a bivalent virus-like particle (VLP) vaccine candidate developed for the prevention of moderate-to-severe acute gastroenteritis caused by norovirus. Originally developed by Takeda Pharmaceutical Company and later licensed to HilleVax, the vaccine consists of VLPs from two norovirus genotypes: GI.1 (Norwalk strain) and GII.4c (a consensus sequence derived from GII.4 variants). It is formulated with an aluminum hydroxide adjuvant and administered via intramuscular injection. The vaccine works by eliciting histo-blood group antigen (HBGA)-blocking antibodies, which interfere with the binding of the norovirus capsid to host cell receptors, thereby preventing viral entry and infection. Clinical development has progressed through Phase 2 trials in various age groups, including infants and adults.
