Hiltonol + SL701 (also known as Poly-ICLC + SL701) is a combination immunotherapy co-developed by Stemline Therapeutics and Oncovir for the treatment of glioblastoma (GBM) and other gliomas. The combination consists of SL701, an off-the-shelf, synthetic multi-peptide cancer vaccine targeting three glioma-associated antigens overexpressed in glioblastoma—interleukin-13 receptor alpha-2 (IL-13Rá2), EphA2, and survivin—and Hiltonol (poly-ICLC), a synthetic double-stranded RNA (dsRNA) that acts as a potent Toll-like receptor 3 (TLR3) agonist and immune adjuvant. By leveraging Hiltonol's ability to activate the innate immune system and enhance antigen presentation, the combination aims to elicit a robust, antigen-specific CD8+ T-cell response against tumor cells. The combination has been evaluated in Phase 1/2 clinical trials for recurrent glioblastoma multiforme.
