Overview

hiNeuroS-TRAIL is an engineered cell therapy consisting of human induced neural stem cells (hiNSCs) that have been developed into a spheroidal format (hiNeuroS) and genetically modified to constitutively secrete the pro-apoptotic protein TRAIL (TNF-related apoptosis-inducing ligand). Developed by researchers at the University of North Carolina at Chapel Hill, this therapeutic platform leverages the innate tumor-homing capabilities of neural stem cells to deliver TRAIL directly to glioblastoma (GBM) sites. TRAIL specifically binds to death receptors on the surface of cancer cells, triggering the extrinsic apoptotic pathway while sparing healthy tissue. Preclinical studies have explored hiNeuroS-TRAIL as a monotherapy and in combination with sensitizing agents like MerTK inhibitors (MRX-2843) or TR-107 to overcome resistance and enhance tumor suppression in aggressive brain cancers.
