Sana Biotechnology's HIP-modified allogeneic induced pluripotent stem cells (HIP iPSCs) are a platform of genetically engineered cells designed to evade the host immune system. The modification involves the depletion of HLA class I and II molecules (via B2M and CIITA knockout) to prevent T-cell recognition and the overexpression of CD47 to inhibit innate immune rejection by macrophages and NK cells. This "hypoimmune" profile allows for the creation of off-the-shelf, allogeneic cell therapies that can survive and function in immunocompetent recipients without the need for systemic immunosuppression. The technology is being applied to various therapeutic areas, including the generation of pancreatic islet cells for Type 1 diabetes and cardiomyocytes for heart failure.
