His-Apo-R15 is a recombinant fusion protein designed for the targeted treatment of malignant melanoma. It consists of a polyhistidine tag (His-tag) for purification, the viral protein apoptin (derived from Chicken Anemia Virus), and a specific RGD-containing peptide (R15) that serves as a targeting ligand. The RGD motif specifically binds to αVβ3 integrins, which are overexpressed on the surface of melanoma cells, facilitating the selective internalization of the protein. Once inside the tumor cell, the apoptin component translocates to the nucleus and induces apoptosis through tumor-specific mechanisms, such as interaction with the anaphase-promoting complex (APC/C), while remaining non-toxic to normal cells like keratinocytes and fibroblasts.
