His6-ZTaq-GS-Cys-DOTA-68Ga is a Gallium-68 (68Ga) radiolabeled affibody conjugate used as a non-specific negative control in preclinical positron emission tomography (PET) imaging studies. The molecule consists of a hexahistidine-tagged (His6) affibody (ZTaq) that specifically binds to Taq DNA polymerase (a non-mammalian protein), a glycine-serine (GS) linker, a C-terminal cysteine (Cys) residue, and the chelator DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) complexed with the positron emitter 68Ga. Because Taq DNA polymerase is not naturally present in mammalian tissues, His6-ZTaq-GS-Cys-DOTA-68Ga does not exhibit specific binding in vivo, making it an ideal control for assessing non-specific uptake, background signal, and biodistribution of target-specific affibody-based PET tracers (such as those targeting HER2 or HER3) in tumor-bearing mouse models.
