Hispolon is a naturally occurring phenolic pigment and polyphenol primarily isolated from the medicinal mushroom species *Phellinus linteus*. It has been extensively investigated for its diverse pharmacological properties, including potent antioxidant, anti-inflammatory, and antineoplastic activities. Mechanistically, hispolon exerts its anticancer effects by inducing apoptosis through the modulation of reactive oxygen species (ROS) levels, activation of Caspase-3, and inhibition of the NF-kappa B signaling pathway. It also demonstrates anti-metastatic potential by suppressing the expression of Matrix metalloproteinase-2 (MMP-2) and Matrix metalloproteinase-9 (MMP-9). Despite its therapeutic promise in treating various cancers such as melanoma, breast cancer, and lung cancer, hispolon's clinical translation is limited by its poor aqueous solubility and low bioavailability. Consequently, current research focuses on developing advanced delivery systems, including cyclodextrin inclusion complexes and liposomal formulations, to enhance its pharmacokinetic profile and chemotherapeutic efficiency.
