Histalean is an oral, betahistine-based small‑molecule drug developed by Obecure as a repurposed therapy for obesity and weight management, including mitigation of antipsychotic‑induced weight gain. It is essentially a new formulation of betahistine, a histamine analogue that acts as an agonist at histamine H1 receptors and a partial antagonist at histamine H3 receptors in the brain, thereby enhancing histaminergic neurotransmission in pathways that regulate satiety, preference for fatty foods, and energy balance. Early Phase II studies in obese adults showed modest but more pronounced weight‑loss effects in women aged 50 years or younger at higher doses, with a safety profile generally consistent with the long clinical history of betahistine in Ménière’s disease.
