**Histone H3.3-K27M neoantigen vaccine** is an investigational therapeutic peptide cancer vaccine developed for newly diagnosed, HLA-A*02-positive diffuse intrinsic pontine glioma harboring the H3.3-K27M mutation. The vaccine presents an H3.3 K27M mutation-derived neoantigen to antigen-presenting cells, with the objective of eliciting mutation-specific CD4-positive and CD8-positive T-cell responses against tumor cells expressing the H3.3-K27M oncohistone. In the Phase 1 ENACTING study, it was administered after biopsy or surgery and conformal radiotherapy, together with the immune adjuvant polyinosinic-polycytidylic acid-poly-L-lysine carboxymethylcellulose. The program has been evaluated by Beijing Tiantan Hospital with involvement of TCRCure Biopharma.
