Overview

HIV-1 broadly neutralizing antibodies (bNAbs) are a specialized class of monoclonal antibodies that target conserved epitopes on the HIV-1 envelope (Env) protein, enabling them to neutralize a wide spectrum of viral variants. These antibodies are being investigated for both the prevention (passive immunization) and treatment of HIV-1 infection. In therapeutic contexts, bNAbs are explored for their potential to maintain viral suppression after the interruption of antiretroviral therapy (ART), a state known as post-intervention control (PIC). They function by blocking viral entry into host cells and can also facilitate the clearance of infected cells through immune effector functions such as antibody-dependent cellular cytotoxicity (ADCC). Clinical development often involves combinations of bNAbs targeting different sites on the Env trimer, such as the CD4 binding site, the V3 glycan supersite, or the V1V2 apex, to prevent viral escape.
