gp140 DP is a recombinant, trimeric HIV-1 envelope glycoprotein (gp140) derived from Clade C strains, developed by Janssen (a Johnson & Johnson company) as a protein subunit component of its heterologous prime-boost HIV vaccine regimens. The drug product is designed to present the HIV envelope in a stabilized trimeric conformation, mimicking the native viral spike to elicit broad binding and neutralizing antibody responses. It was evaluated in major clinical trials, including the Phase 3 Mosaico (HVTN 706) and Phase 2b Imbokodo (HVTN 705) studies, where it was administered as a boost following priming with adenovirus serotype 26 (Ad26) mosaic vectors. However, these large-scale efficacy trials were terminated in 2021 and 2023 after the regimens failed to provide significant protection against HIV-1 infection.
