HIV-1 DNA pGP140 is a DNA plasmid vaccine candidate developed by the National Center for STD and AIDS Prevention and Control at the Chinese Center for Disease Control and Prevention (China CDC). It is designed to express the HIV-1 envelope glycoprotein gp140, a key target for neutralizing antibodies. In clinical development, it is typically used as a priming agent in a heterologous prime-boost immunization strategy, often co-administered with another DNA plasmid, pGPNEF (expressing Gag, Pol, and Nef), and followed by a boost with a recombinant vaccinia virus (TianTan strain) expressing HIV-1 antigens. This approach aims to stimulate both cellular (T-cell) and humoral (B-cell) immune responses to prevent HIV-1 infection. The vaccine has been evaluated in Phase I and Phase II clinical trials in China, exploring various immunization intervals and the use of electroporation to enhance DNA delivery and immunogenicity.
