Overview

The HIV-1 recombinant Ad5 vaccine is a viral vector-based vaccine designed to prevent HIV-1 infection and control viral replication. It utilizes a replication-defective adenovirus type 5 (Ad5) vector to deliver genetic material encoding specific HIV-1 proteins, namely Gag, Pol, and Nef, from HIV-1 clade B strains. The vaccine aims to elicit cell-mediated immune responses, particularly T-cell immunity, against these HIV-1 antigens. Developed by Merck & Co., this vaccine was notably evaluated in the Step Study (HVTN 502) and Phambili (HVTN 503) clinical trials. However, these trials were terminated due to the vaccine's failure to prevent HIV acquisition and, in certain subgroups, an observed increase in the risk of HIV infection, particularly in uncircumcised men with pre-existing Ad5 immunity.
