HIV-1 Trimer 6931 is a synthetic, soluble, prefusion-stabilized trimeric form of the HIV-1 envelope glycoprotein (Env), designed as a prophylactic vaccine candidate for the prevention of HIV infection. It is based on the ConC_Base0 variant and incorporates multiple stabilizing mutations—including three at the trimer apex (302M, 320L, and 329P), two glycine substitutions in gp41 to improve stability and antigenicity in its prefusion state, standard SOSIP substitutions, and seven additional stabilizing mutations. The vaccine is administered with an adjuvant (Adjuplex) to enhance immunogenicity. Its mechanism centers on stimulating immune responses—specifically inducing broadly neutralizing antibodies against diverse tier 2 primary isolates of HIV by focusing immune responses on conserved epitopes such as the fusion peptide region. Developed by the National Institute of Allergy & Infectious Diseases (NIAID), it has been evaluated in phase I clinical trials both alone and as part of prime/boost regimens with other Env-based vaccines[5][3][6].
