Overview

The HIV E-vaccine is a prophylactic and therapeutic vaccine candidate developed by Covalent Bioscience, with plans for further development under a spin-out entity, E-Vaccines Inc. (EVI). The vaccine utilizes a novel electrophilic immunogen approach to target the highly conserved CD4 binding site (specifically the 421-433 region) of the HIV gp120 envelope protein, which is essential for viral entry into host cells. Unlike traditional vaccines, the E-vaccine employs electrophilic analogs to induce the production of catalytic antibodies (catabodies) and irreversible antibodies capable of neutralizing genetically diverse HIV strains. This strategy is designed to bypass the defective IgM-to-IgG class switching that typically hinders effective antibody responses to this conserved region, thereby generating mature, broadly neutralizing IgG antibodies. The technology is based on over 30 years of research and has been supported by NIH grants, though the developer Covalent Bioscience has reportedly ceased operations.
